I receive Prolotherapy treatments every 4 weeks or so for the past 2 months on my infraspinatus/rear deltoid. I last received therapy 2 days ago. Today I went to the gym and did some weight training using about 40% my normal lifting weight. My shoulder feels very sore and I feel heat and inflammation. Is this normal? And if I did go too hard/too fast is this going to negate the further healing effect and properties of the prolo treatment?

Well not to scold you, but this would have been a great question to ask while getting the treatment.
Are you aware of the mechanism of action of prolotherapy? It is intended to re-initiate the inflammatory process which is in line with your subjective complaints of soreness and inflammed.
Do I anticipate exercising at 40% intensity as being detrimental to the treatment, no. But calling and finding out would be the best action. With steroid injection they tend to limit activity for 24 hr, doubt there is any more restriction with prolo but I would find out by calling.
