Starting around month 3 that shit had me feeling like I was on cocaine. I literally had to be doing something that involved moving my body at all times. Like I couldnāt sit still. That lasted a few months or so and then lessened or either it became the new normal.
Im 5 weeks in and feel nothing so far, maybe a slight increase in Joie de vivre. Just started max dose this AM.
- 3 weeks at 150mg
- 2 weeks at 300mg
Week 6 started 450mg

She started Sept 4. I do believe that should give 8 weeks.
Just over two weeks now at max dosage of 450mg (extended release). So far I have noticed nothing except elevated heart rate (up about 5 bpm) and extended time to sleep (up from 2 mins to 10-15mins). Seven weeks now since starting Bupropion.
Wife just switched to the 300 one XR.
I didnāt get much from wellbutrin but a little extra energy.
Iād run it a few more weeks at that those and then discontinue it if nothing. I went cold turkey off it and felt nothing.
Yeah the doc said he wasnāt hopeful with this drug, for whatever reason. I know @dextermorgan said you need to get the actual Wellbutrin product but its extremely cost prohibitive. Perhaps I am missing the Cytochrome P450 2D6 (CYP2D6) enzyme like hardartery mentioned.
Doc said the next step would be modafinil. @hardartery is that enzyme required for modafinil as well?

So, I am exploring alternatives. I am trying phenylalanine right now, it is supposed to be be able to be processed by two separate pthways, one of which bypasses the CYP2D6 path, but I donāt think that itās doing much for me. I am still exploring.
Keep me posted. Maybe my psychiatrist will order a Psychotropic Test.

I will look into it. Iāve heard of piractam via a podcast I think. Ive never heard of this analog thoughā¦based on that website there are a LOT I havent heard of jesus. Im going to give the bupropion a bit longer as just this past weekend I was having some clarity of thoughts and speech for the first time in years. Hopefully it is ramping up in my system.
Six weeks now at 450mg bupropion and only a slight improvement and only in short term memory and concentration. Doc is going to drop my bupropion dose down to 300mg and add in modafinil @ 100mg to start. Touching base in another 4 weeks. He mentioned a bunch of ADD drugs we could try as well but trying this first.
To note, resting heart rate went from 52 to 60 and BP went from 116/64 to 134/70.
What ADD drugs have you ever used successfully? Iām going to make a modafinil prediction for you, itās āLOLā. Good luck with that gem working for ADD. Not trying to be negative, just sayingā¦
Iāve never used ANY because Iāve never been diagnosed with ADD. Even now the doc is just trying to get me some wakefulness and concentration. The bupropion helped a tad but nowhere near what others have reported. So itās different than your case, I think, but we may still be similar genetically.
Regarding the Cytochrome P450 2D6 (CYP2D6) enzyme we talked about, are there varying degrees of its presence? Might I have 20% or less of what is considered normal or is this an all or nothing type thing?

There are varying degrees, for sure. Even within apparently identical mutations. They are doing a lot of work on this right now, and identifying hundreds of separate mutations and sub-mutations. We literally have no idea how much we donāt know on genes and mutations right now. This is probably the most heavily researched cytochrome, and they are just scratching the surface.
Nothing noticeable at all at 100mg daily.
Zero effect from Modafinil so doc wants to try Adderral. So far the bupropion is the only thing having any effect and its pretty minimal. I will say however going from 450mg to 300mg bupropion I noticed a difference.
Im willing to bet hardartery is right here and effects will be minimal on new drug. Doc said with me 5mg would be a waste of time so starting me at 10mg. Keeping bupropion at 300mg for a few days to see how I am and if ok then he said I can go back to 450mg.
Memory seems a bit better these days and the fog has mostly lifted but I have ups and downs. I am still just closing-my-eyes-for-30-seconds away from falling asleep at any moment. Luckily my most wakeful time is first thing in the AM so the gym has been good and my strength gains are coming back to where I was in march.
Keeping T steady at 120mg a week via two shots. Havent had bloods done in a while so may get some soon. Im guessing my trough TT is around 1200 and FT around 35.
