Freezing hCG Question

Greetings,
So a week ago i reconstituted some hcg with 0.9% NaCl water and pulled it out in individual insulin syringes containing 750 iu planning to take 750 iu 3x per week.

I pinned 750 iu and froze the remaining syringes. Next day i felt a good libido boost and a feeling of well-being which i usually get from hcg.

But the problem is that i didnt feel anything after subsequent shots.

Did i destroy the hcg by freezing?

And lets say i want to test my hcg using pregnancy test. How long does the hcg stay in your system?
I know that monday shot was good and i dont know wether that one might flag the pregnancy test still?
I want to see whether the frozen hcg (once thawed and pinned immediately) still has potency.

What do you all think?

It will lose some potency by freezing which I’ve done in the past, but for long term storage (more than 90 days) its better than refrigeration.

Just to be clear, you don’t pee yourself on the pregnancy test to check the efficacy of the frozen hcg. You thaw a frozen hcg syringe and then eject the hcg itself onto the pregnancy test.

Freezing is part of the lyophilization process hCG undergoes, so it stands to reason it won’t hurt potency especially since it is only once.

yes, only one cycle of freezing/thawing.

i have read conflicting info on freezing hcg, that is why i asked.

a freeze thaw cycle should be fine, loss of potency should be accetable. Multiple cycles are quite likely problematic.

Lyophilization and freezing share the same first step, thats correct. However, lyophilization uses shock freezing at temp of -80 to -170°C wheras in your case i guess you freeze at -20. Generally speaking, the slower the freezing process the more harm the protein will experience. This has to do with very harsh conditions at the boarder between the liquid and the solid state, which harms the protein.

Using a pregnancy test to check for a potency loss is absolutely meaningless. 1) its a qualitative and not quantitative readout and 2) the sensitivity of the test is waaaaay to high. A pregnancy test has a typical sensitivity of 50 IU/L. So even if you dissolve eg your 5000 IU vial in 1L of a liquid solution, then even if you loose 90% of your hcg potency the test woukd still show a big fat positive result.

There is conflicting information everywhere. A guy on a domesric forum over here swears that hcg always loses potency (becomes useless) even if mixed with bacteriostatic or saline water and kept in the fridge after only 48 hours.

I find that kinda hard to believe as i always thought it was good for 30-60 days in the fridge.

However i chose to freeze it because i mixed it with saline water containing no benzyl alcohol.

Perhaps the loss of libido can be explained by changing T:E2 levels. Let’s stick to Occam’s Razor.
